Impacto del Coenzima Q10 (Ubiquinol) en el compromiso del endotelio vascular cerebral en la enfermedad de Alzheimer: Valoración funcional y metabólica in vivo e in vitro.
La disfunción vascular puede contribuir al desarrollo y progresión de la enfermedad de Alzheimer (EA), según datos epidemiológicos y experimentales. Existen numerosas anormalidades estructurales y funcionales de la microvasculatura en estos pacientes. El péptido ß-Amiloide (Aß) daña el endotelio induciendo la generación de especies reactivas de oxígeno mediada por la activación de la NADPH oxidasa y por su incorporación en la mitocondria originando disfunción mitocondrial. En esta línea las bases moleculares y celulares de la protección por el CoQ en células endoteliales cultivadas frente al Aß, su efecto en la funcionalidad in vivo de la microvasculatura en el modelo murino de Alzheimer 3xTg-Ad y colaboramos con el Servicio de Neurología del HGUCR para para objetivar in vivo si la suplementación oral con CoQ (ubiquinol) o placebo mejora la situación funcional del endotelio vascular cerebral en un grupo de pacientes con criterios clínicos de deterioro cognitivo leve midiendo la vasoreactividad con Doppler transcraneal.
Modulación del estrés oxidativo en glioblastoma multiforme y su reprogramación metabólica.
El glioblastoma multiforme es el tumor del SNC más frecuente y con peor pronóstico. Estamos estudiando en modelos in vitro e in vivo como el tratamiento previo con coenzima Q remodela el metabolismo del tumor y el proteoma, previniendo la glucólisis aeróbica (efecto de Warburg), que tiene un impacto inmediato en el crecimiento y la invasión del tumor produciendo una disminución de la viabilidad de las células de glioblastoma in vitro y triplica el efecto de la radioterapia a corto y largo plazo y el efecto proapoptótico de la temozolomida.
Actualmente estamos comenzando un ensayo clínico en el que participamos con varios Servicios del HGUCR para evaluar el impacto pronóstico de la suplementación con el complemento dietético ubiquinol en el tratamiento de pacientes con glioblastomas primarios según el protocolo adyuvante Stupp
Papel neuroprotector del conezima Q10 en la proteostasis durante la enfermedad de Alzheimer
Un aspecto común a las enfermedades neurodegenerativas es el incremento en la producción de especies reactivas de oxigeno (ROS) por una disfunción mitocondrial. El incremento de ROS altera la proteostasis celular por el desbalance entre la respuesta a proteínas mal plegadas (UPR), degradación de proteínas por vía proteasomal (ERAD) y autofagia, conduciendo a la vulnerabilidad y muerte celular por apoptosis. Estamos estudiando la interrelación entre el estrés oxidativo y la proteostasis, estableciéndose su dinámica espacio-temporal, en modelos in vitro e in vivo y la regulación temprana del nivel de estrés oxidativo con un suplemento de ubiquinol (CoQ) en etapas prodrómicas y premórbidas de la enfermedad, para controlar la proteostasis celular y así ralentizar el progreso de la patofisiología asociada a la enfermedad de Alzheimer en el modelo murino 3xTg-AD de esta enfermedad.
